Partial Sequence Analysis of Hibiscus syriacus cDNA Clones

|
To study the genome of Hibiscus syriacus, we generated 110 expressed sequence tags (ESTs) from cDNA leaf library. A homology search of these 110 ESTs against the NCBI (National Center for Biotechnology) non-redundant protein database revealed that 19 clones (17.3%) were significantly similar to the protein coding sequences. Nine clones had relatively high homologies to retrovirus-related pol protein genes. Ten c DNAs had high degree of sequence similarities when compared with genes from other organisms. The remaining cDNA clones seem to be new genes not only in Hibiscus syriacus genome but also in all organisms. The average size of insert was about 2.66 kb (SD = 1.66 kb). In this paper, we also report an efficient and rapid method for RNA isolation from high polysaccharide-containing material, like Hibiscus syriacus.
